A new study from New York University and the University of Rochester suggests that a new type of drug, pembrolizumab, could help to reduce the risk of Alzheimer’s disease more profoundly. In mice that have inherited the Aβ-amyloid receptor, which is thought to contribute to the illness, the drug reduced the incidence of the disease by 75%. It also reduced the frequency of cognitive impairments that are characteristic of Alzheimer’s by 70%.
Mice with familial Alzheimer’s disease (FAD) have a mutation in the gene for the receptor. The Aβ peptide found in the blood and brain is the key toxic compound that causes the disease. The Aβ protein and other Aβ peptides can’t migrate from the brain into other cells. This leads to the brain accumulating the protein in plaques, which are a hallmark of Alzheimer’s.
The new report finds that the pembrolizumab, developed by the biotech company Genentech, was able to prevent the deposition of the protein — which is typically destroyed by chemotherapy, which causes the production of Aβ particles. The drug was also ineffective when an enzyme that breaks down the protein was used instead.
The researchers also explored whether the drug could be used to treat neurodegenerative diseases that can cause the buildup of the Aβ protein. The results indicate that a genetic defect in the receptor, which they call “FAD-like genetic defects of Aβ,” could prevent the deposition of the protein from the brain into other organs.
While pembrolizumab may not cure Alzheimer’s, it does appear to be promising in helping to slow its progression and potentially treat certain types of the disease.
The study was funded by grants from the National Institutes of Health, the National Eye Institute, the National Institute on Aging, and the U.S.-based Gilead Sciences Foundation, which also funded other work in the group.
